Phưngphápchếtạothuốcdánktoprofn:dùngđểchếtạothuốcđángồmDurotakvàketprofen.Ketoprofenđược phântánvàtrộnđềutrongđungmôihữucơchứaDuro­Tak®87­2852,MentholvàBHT.Hỗhọpsaukhiloạicácbọt khíđượctràiiênlóplưngkhôngthấmbằngpolyester.Lớpcốtthuốcđượclàmkhôtrong 10phútờnhiệtđộphòngvà1 giờờnhiệtđộdưới50°c. Địnhlượngktoprofn: Ketoprofen trong thuốc đán được định lượng bằng sắc ký lỏng hiệu năng cao. Lượng ketoprofenchứaIrongthuốcdánđượctínhtrênlượngchứatrong1đơnvidiệntích. Thửđộhòatan:Lượngkeioprofenhòatanđượcxácđịnhbằngcáchdùngbộdụngcụsố5,phépthử3củachuyên luậnthuốcdánDượcđiểnHoaKỳ.Lươngketoprofenhòatanđượcxácđịnhbằngsắckýlỏnghiệunăngcao. Kếtquả: Chếtạothuốcdánktoprofn:ĐồnglượngĐurotak®87­2852vàMentholđuợcbỏvào i bnh.Hỗnhợpđượctrộn đềutrong 3 phútđể tạo cốt thuốc đồng nhất. Kctoprofenđược trộn đều trongcốtthuốc Đuro­Tak®87­2852. Butyl hyđroxytolulenecũngđượctrộnđềutrongcốtthuốcketoprofen/Durotak.cốtIhuốcđượctràilênlớplưngkhồngthấm đạtkhốilượngxấpxỉ 30±3 mg/70cm2saukhisấykhô.Lácốtthuốcketoprofen/Duro­Tak®đượccắtthànhhnh dạngvàkíchthướcmongmuốn.Đónggóivàobaob(lánhôm61 m)chốngthấmẩmvàkhôngkhí.Bảoquànởnhiệt độkhôngquá25°c Địnhlượngktoprofn:mỗiláthuốcđánchứa30±3mgketoprofenchokíchthuớc70cm2. Thửđộhòatan:lượngketoprofenphóngthíchtừthuốcdánKetoprofentuyếntínhtheotVi. Preparationfortransdermaỉpatchescontaining30mgofKetoprofen Summary KetoprofentransdermapatchusingDuro­Tak®adhesiveswasdevelopedtodeliverprecisequantitiesofketoprofen totheskinforaprolongedperiodoftime.Duro­Tak®87­2852wasmixedwithmentholinordertomakeanadhesive which is highly suitable for long­term wear on human skin but also can be removed without causing irritation or damagetotheskin. Materialsandmethod: Matrials:Duro­Tak®87­2852,Menthol,KetoprofenandButylatedhydroxytoluene. Prparation ofKtoprofnpatch: Ketoprofen was dispersed and evenly mixed with organic solvents contained Duro­Tak®87­2852,MentholandButylatedhydroxytoluene.Afterremovingairbubbles, themixturewascastedona pieceofimpermeablepolyesterbackingfilm. Dtrmination of ktoprofn contnt: The Ketoprofen patch was analyzed by high performance liquid ,chromatographicmethod.Thetotalamountofnicotinedividedbytheareaoftheketoprofencontentperunitarea. ỉn vitro rlas studis: Theketoprofen released from thepatch was determined by using the ƯSP Dissolution Apparatus 5, thestainless steel diskassemblybeingreplacedwitha watchglass (TheDrugReleaseTest3ofƯSP TransdermalSystem).TheamountofketoprofenwasanalyzedbyHPLCmethodaspreviouslydescribed. Results: PrparationofKtoprofnpatch:A50/50blend(w/wbysolids)mixtureofDuro­Tak®87­2852andMentholwaspoured intothesamejar.Thewetblendwasmixedwiththelaboratorypropellermixerfor3minutestocreateauniformmixture. KetoprofenwashomogeneouslyblendedthroughoutthemixtureofDuro­Tak®87­2852.Butylhydroxytohilenewas then mixed togetherwith the ketoprofen/Duratak adhesive matrix. Thewetblendwas coatedonto an impermeable polyesterbackinglayertoachieveapproximately30± 3mg/70cm2drycoatweight. Thelaminiatcdketoprofen/Duro­Tak® sheetmaythenbecutintoanydesired shapes andsizes, packagedinto moistureandoxygenimpermeablepackingmaterial(aluminiumfoil)andkeptatnotmorethan25°cinarefrigerator. Dtrminationofnicotincontnt:EachKetoprofenpatchcontains30±3mgofketoprofenwiththesizeof70cm2. Invitrorlasstudis:dieamountofketoprofenreleasedfromKetoprofenpatchesshowsastraightline. Conclusion: Amedicalgradeadhesivecomprisesanacrylatevinylacetateself­curingpressuresensitiveadhesive(Duro­Tak®87­ 2852)wasusedlomadeKetoprofenpatches. The Ketoprofenpatches wereevaluated fortheircontent, ketoprofen release. IV.KẾTLUẬN ­ĐãphântíchbằngDSC,DTA,phổFTIRvàđánhgiácáctínhchấtlýhoácủathuốcdán. ­Đãnghiêncứusàngỉọcchếtạocácthuốcdángồm1lớphaynhiềulópcốtthuốccóđộdàythayđổitừ 25­ 1 2 5 ^m.Đãxác đ ị n h đượccôngthứcphùhọpnhất:thuốcdánchứa30mgketoprofentrênđiệntích70 cm2,cốithuốcnặng700mg,độdày100^m.Nồngđộketoprofenvàmentolđồnglượnglà4,28%. 737
8. ­Đãchọnđượccôngthứctổtnhấtgồmketoprofen,Durotak87­2852và1­mentol.Trongđó,tỷlệphối hợp ketoprofen, 1­menthol là 1:1 có sựtương tác tốtnhấtnên đượcgiữổn định thuốcdan, độ hoatan ketoprofentốtnhất. ­Đãxâydựngquytrnhchếtạothuốcđánketoprofenổnđịnhbằngphươngphápdungmôihữucơ,loại1 lớptrongsuốt.Khốilượng,hàmlượngketoprofenvàI­mentholđềuđạttronggiớihạn90­110% sovớihàm lượngghtrênnhãn. ­ĐãxâydựngđượcphươngphápđịnhtínhvàđịnhĩượngketoprofenbằngHPLC. ­Đãxâydựngđiềukiệnthửnghiệmphóngthíchketoprofentừthuốcđándựatheocácquỳđịnhchung củaƯSP30.Tốcđộphóngthíchcủathuốcđánketoprofen(độđày100|xm,chứa30mgketoprofen/70cm2) là22,06|ig/cm2/giờvớihệsốtươngquan0,98.Hệsốkhuếchtán32,58. 10‘10cm2/giây. ­ĐẵxácđịnhthuốcdánKetoprofenkhônggâykíchứng.
